Lead Compound

A lead compound is a chemical that shows promise as a drug but is not ready for testing in humans. It has activity against a biological target, but it may also have poor solubility, rapid metabolism, or off-target effects. The lead is a starting point, not a finished product. Medicinal chemists take the lead and modify its structure, adding or removing chemical groups to improve potency, selectivity, and safety. This process, called lead optimization, can take years and produce hundreds of variants before a candidate is selected.

Leads come from many sources. High-throughput screening of compound libraries can identify hits that become leads. Natural products, like plant extracts or bacterial fermentation broths, have yielded many leads. Rational design starts with the structure of the target protein and builds a molecule to fit. Fragment-based approaches piece together small molecular fragments that bind to different parts of the target. Once a lead is identified, it enters a cycle of design, synthesis, testing, and refinement.

What makes a good lead:

Most leads fail. A lead that looks good in a test tube may not work in cells, and one that works in cells may not work in animals.
